Interleucina-10 y su Dualidad en el Cáncer de Hígado: Un Estudio Estructural y Computacional sobre su Regulación de la Apoptosis

La interleucina-10 humana (IL-10) es una citocina antiinflamatoria con un papel complejo en la apoptosis y proliferación celular, especialmente relevante en células HepG2 de cáncer de hígado. Su acción pleiotrópica se media a través del receptor IL-10R1/R2, capaz de activar vías de señalización tanto pro-supervivencia como pro-apoptóticas, dependiendo del contexto celular. El dominio 1 en su hélice A dentro de la estructura 3D de la IL-10 se identifica como un sitio clave para su interacción molecular. El objetivo de esta investigación fue la caracterización mecánico cuántica de la hélice A mediante la Teoría del Funcional de la Densidad (DFT), calculando descriptores globales de reactividad (DGR) y los orbitales moleculares de frontera HOMO-LUMO. El análisis computacional proporcionó evidencia fundamental sobre las propiedades electrónicas y la reactividad de esta hélice, vinculándolo con su función biológica en la homeostasis y apoptosis celular. Modelamos y caracterizamos la hélice A del dominio 1 de IL-10 humana, correlacionando nuestros resultados con su contexto biológico reportado por experimentos in vitro e in vivo en bases de datos científicas. Los resultados, que incluyen la identificación de sitios de reactividad molecular, son cruciales para el diseño racional de moduladores de la IL-10 y péptidos miméticos. Este enfoque representa una estrategia terapéutica potencial dirigida contra el carcinoma hepatocelular. Nuestra investigación sugiere que la integración de métodos computacionales avanzados, como la DFT, junto con su validación experimental, es fundamental para esclarecer el papel dual de las citocinas en la progresión del cáncer y para impulsar el desarrollo de nuevas estrategias terapéuticas.
